Description

The aim envisioned in this interdisciplinary project is experimental optical detection and quantification of the activity of the enzymes responsible for mRNA degradation using novel double-labelled nucleotide and mRNA-based fluorescent probes. The optical response of those probes can be strongly enhanced if chemically grafted to plasmonically active silver nanowires. Therefore, careful control of the surface modification of the plasmonic particles is required so that the distance between the particle and the probe is adjusted to ensure efficient enhancement of the fluorescence. The approach with plasmonic particles should give the opportunity to monitor the reaction at a single-molecules level and the activity in real time.

Key responsibilities include

      The post-doc will be responsible for performing fluorescence microscopy experiments for single- and double-labelled probes conjugated with silver nanowires in liquid environment with varied excitation sources and detection schemes. Designing appropriate modifications of fluorescence microscopes in order to investigate enzyme activity using the FRET effect in real-time arrangement.

Resposibilities:

  • to modify the surface of silver nanowires with labelled nucleotide- and mRNA-based molecular probes
  • to study influence of plasmon excitations in silver nanowires on the optical properties of molecular probes
  • to fabricate reaction cells
  • to investigate of enzymatic activity for double-labeled probes densely packed on silver nanowires  
  • to investigate of enzymatic activity for individual FRET probes
  • to disseminate of research results at project meetings, in peer-reviewed journals and at international conferences

We offer a post-doctoral position (full-time employment) in the Institute of Physical Chemistry PAS with a gross salary of Around 9650PLN gross per month financed from the NCN OPUS 25 2023/49/B/ST4/01450 „Plasmonic nanoparticles decorated with fluorescently-labelled mRNA: using singlepair FRET effect for investigating enzymes active in mRNA metabolism”  

100% of the work time of Postdoc will be devoted to the realization of the proposed project. The offer is for    12 months with a possible extension

Career development prospects at IPC PAS

The successful candidate will gain experience in optical detection and quantification of the enzyme activity responsible for mRNA degradation which may influence metabolism, gene expression, signalling and etc. The investigation of the photonic control of light-matter interaction at the nanoscale can be beneficial in construction of new hybrid systems based on RNA.

The successful candidate will be eligible for all programs at the IPC PAS, including the IPC PAS Young Scientists competition and the IPC PAS Young Scientists Mobility competition.

Requirements

Completed PhD degree in chemistry, physics or related fields meeting the requirements https://www.ncn.gov.pl/sites/default/files/pliki/uchwaly-rady/2023/uchwala23_2023-zal1_ang.pdf;

Knowledge of physical chemistry, nanomaterials, plasmonics, surface modification, molecular interactions, fluorescence microscopy;

Experience in operating optical fluorescent microscopes;

Experience in analysis of fluorescence images and data;

Experience in work in research grants;

A strong motivation for experimental interdisciplinary work

Key assessment criteria

The Commission will take into account the following criteria and evaluate applications on a point scale:

•  the candidate’s research achievements, including publications in academic journals

•  research-related achievements, scholarships, awards and research experience, research workshops and training courses, participation in research projects

•  the candidate’s competence to carry out specific tasks in the research project

Candidate must meet formal requirements:

a) the project leader was not the supervisor or co-supervisor of the candidate doctoral dissertation;

b) obtained a doctoral degree in an entity other than the entity where employment in this position is planned, or completed at least 10 months of continuous and documented postdoctoral internship in an entity other than the entity implementing the project and in a country other than the country of obtaining the doctoral degree;

c) will be employed for a period of no less than 6 months;

d) during the period of receiving this remuneration will not receive any other remuneration from funds granted as direct costs from research projects financed under NCN competitions;

e) during the period of receiving this remuneration will not receive remuneration from another employer on the basis of an employment contract, including an employer based outside Poland;

f) during the period of receiving this remuneration will not receive retirement benefits from the social insurance system.

Postdoc

     A post-doc type post is a full-time post, scheduled by the project’s principal investigator for a person who has been conferred a PhD degree in the year of employment in the project or within 7 years before 1 January of the year of employment in the project. For researchers with multiple doctoral degrees, the reference date is the date of the first award of the doctoral degree. This period may be extended by a time of long-term (in excess of 90 days) documented sick leaves or rehabilitation leaves granted on account of being unfit to work. In addition, the period may be extended by the number of months of a childcare leave granted pursuant to the Labour Code and in the case of women, by 18 months for every child born or adopted, whichever manner of accounting for career breaks is preferable.
