The intricate nature of intrinsically disordered proteins involved in gene silencing - and how to deal with it

Intrinsically disordered proteins (IDPs) are an important class of biomolecules that regulate biological processes in higher organisms. Their lack of fixed spatial structure facilitates their regulatory functions and enables the efficiency of biochemical reactions to be controlled by temperature and the cellular environment. From a biophysical perspective, IDPs are biopolymers with a wide configuration state space and their actual conformation depends on the non-covalent interactions of their amino acid side chains at a given temperature and chemical conditions.
I will discuss the properties and interactions of an intrinsically disordered GW182 protein that interacts with the pivotal CNOT1 subunit of the CCR4–NOT deadenylase complex, thereby mediating microRNA-dependent gene silencing. The CCR4–NOT complex also functions in an alternative gene silencing pathway that is regulated by an intrinsically disordered tristetraprolin (TTP) protein. While the CNOT1 residues involved in the specific TTP binding have been identified, the molecular mechanism underlying the CNOT1 interaction with GW182 remains unknown. We use a combination of hydrogen-deuterium mass spectrometry, fluorescence correlation spectroscopy, and the liquid-liquid phase separation (LLPS) propensity of the GW182 silencing domain (SD) to identify the molecular determinants of CNOT1 - GW182 recognition. We demonstrate that the GW182 silencing domain undergoes LLPS with a lower critical temperature due to a tryptophan-dependent LLPS mechanism. We also show that the GW182 SD forms two-component condensates with the CNOT1 protein via a host-client relationship, and that this process can be inhibited by single point mutations in the CNOT1 binding site or by competition with TTP peptides for the binding site within the ternary protein system. To verify the specificity of protein interactions within the dense liquid phase, we combine Förster resonance energy transfer (FRET) with fluorescence recovery after photobleaching (FRAP). Thanks to its angstrom spatial resolution, FRET-FRAP allows us to distinguish between proteins that are specifically involved in complexes and those that have been accidentally trapped in microdroplets due to non-specific polymer interactions. This approach provides a generalisable, selective framework for studying protein complexes in crowded molecular environments.

Decoding the biological "languages" underlying genetic and cellular states remains a major challenge. Single-cell omics measurements are transforming our understanding of biology; however, they are expensive and destructive, making it difficult to monitor live cells in tissues and humans over time. By contrast, imaging is non-destructive, low-cost, and scalable, but it can be difficult to interpret.
My lab focuses on developing generalizable and scalable experimental and computational frameworks that leverage generative AI to bridge the gap between diverse biological data modalities in order to decode, model, and reprogram cells in multicellular systems. For example, we have developed technologies that (i) predict single-cell omics data from non-destructive imaging modalities, (ii) reconstruct molecular dynamics in live cells over time using imaging, and (iii) generate cellular and tissue images directly from single-cell gene expression profiles. These approaches enable rapid and scalable querying and prediction of multi-omics and imaging information in real time, in live cells, across both mammalian and microbial systems.
By integrating multi-modal live-cell and label-free imaging with genetic and chemical perturbations and engineering, we have created massively scalable screening systems to investigate cell functions and behaviors across space and time, with the ultimate goal of enabling the de novo design of cells using AI.
Translating these distinct "languages" (data modalities) of biology unifies different perspectives of cell and tissue biology, reduces the need for multiple destructive measurements, and moves us closer to our long-term vision: building digital simulators to read, write, and engineer multicellular systems.
Biogram:
Dr. Jian Shu (https://jianshulab.org) is an Assistant Professor at Harvard Medical School and Massachusetts General Hospital, Harvard-MIT Health Sciences and Technology, as well as an Associate Member at the Broad Institute of MIT and Harvard. Dr. Shu was the valedictorian at Peking University, where he studied stem cell biology and cellular reprogramming with Dr. Hongkui Deng. He was a Helen Hay Whitney Fellow, working at the intersection of single-cell genomics and stem cell biology in Dr. Eric Lander’s lab at the Broad Institute of MIT and Harvard, as well as Dr. Rudolf Jaenisch’s lab at the Whitehead Institute for Biomedical Research at MIT.
Dr. Jian Shu joined the faculty at MGH/Harvard Medical School and established his lab in January 2021. The Shu Lab's research focuses on developing scalable experimental and computational methods to decode and translate the various 'languages' (data modalities) of biology across different scales—from molecules to cells, tissues, and organisms. These methods aim to unify different perspectives and maps of cell and tissue biology, significantly reducing the need for multiple measurements and shifting biology from direct measurement to prediction and generation through AI. The long-term goal is to build digital twins of multicellular systems and virtual simulators of living systems.
Dr. Shu has received awards and grants from the NIH New Innovator Award DP2, the Pathway to Independence Award K99/R00, the NIH Common Fund Cellular Senescence Network UG3/UH3, NIAID P01, multiple R01s, MLSC, the Burroughs Wellcome Foundation, Additional Ventures SVRF, Additional Ventures Expansion Award, the Leo Foundation, the Harvard Stem Cell Institute, MIT-MGB HEALS and SPARC at the Broad Institute.

This year marks the 130th anniversary of the establishment of the Nobel Prize, by the last will of its founder, Alfred Nobel (1895). In keeping with Nobel tradition, this year's prizes will be officially presented in Stockholm on December 10, 2025, at prestigious ceremony organized by the Royal Swedish Academy of Sciences.
This year, the Nobel Committee awarded with the prize in chemistry three scientists: Professor Richard Robson from Australia (born 1937, United Kingdom), Professor Susumu Kitagawa from Japan (born 1951, Japan), and Professor Omar M. Yaghi from the United States (born 1965, Jordan). The Nobel Committee divided the prize equally among the three laureates, adding an intriguingly brief justification: "for the development of metal-organic frameworks".[1]
This achievement can be considered as another triumph of coordination chemistry, however a closer look allows us to appreciate the multidisciplinary nature of this achievement. At this point, it is worth emphasizing the essence of the definition of MOFs and the fundamental breakthroughs related to porosity of materials.

The chemistry of transition metals continues to surprise us when pushed to its limits of oxidation, bonding, and composition. Using first-principles density-functional theory and evolutionary algorithms for crystal-structure prediction, we will explore those extremes—from the hypothetical Ni⁵⁺ oxidation state in oxygen-rich nickel oxides to unusual Pd–Pd bonding and subvalent silver halides. Our calculations reveal why oxygen-saturated nickel oxides beyond NiO₂ are intrinsically unstable: excess oxygen drives reduction of nickel and the formation of superoxide or peroxide species instead. In contrast, palladium and silver exhibit remarkable tendencies toward metal–metal interactions, leading to multicenter Pd(I)–Pd(I) bonds in predicted Pd₂O structures and to rare Ag₆ clusters with localized two-electron–six-center bonding in Ag₆Cl₄. Together these results chart the energetic and structural frontiers of inorganic chemistry, highlighting how computation can uncover compounds that test the boundaries of oxidation, reduction, and metallic bonding—materials that may be difficult to stabilize experimentally but that expand our understanding of how elements behave under chemical extremes.

Interferometric near-infrared spectroscopy (iNIRS) is an optical technique for non-invasive monitoring of cerebral blood flow in humans. In this seminar, the speaker will present recent progress in iNIRS, covering both its technical aspects and its practical use. Applications will be illustrated with examples of recent experimental results in humans.
Biography
Marcin Marzejon, Ph.D., is an optical engineer specializing in biomedical optics and imaging. He earned his Ph.D. summa cum laude in Automation, Electronics, Electrical Engineering, and Space Technologies from Gdańsk University of Technology in 2023, with pioneering work on two-photon vision.
Dr Marzejon is currently a Specialist at the Institute of Physical Chemistry of the Polish Academy of Sciences, where he develops advanced interferometric near-infrared spectroscopy techniques for non-invasive bioimaging. He has co-authored nine peer-reviewed papers, sixteen conference presentations, and holds two patents. His expertise spans fiber-optic sensors, Raman spectroscopy, two-photon vision and imaging, and quantitative phase imaging, alongside strong skills in software development, data analysis, and system prototyping.

- Abstract:
The aim of this seminar is to present a concept for optimizing the design of ZrC protective coatings for biomedical applications. The coatings were deposited using the PVD (Physical Vapor Deposition) technique on 316L medical steel substrates. The goal of the optimization was to develop a protective coating structure and geometry that would achieve increased fracture resistance, low frictional wear, high adhesion to the substrate, high corrosion resistance, and antimicrobial properties. For this purpose, seven decision criteria were proposed based on the internal stress states in the coatings and the distribution of strains initiated by external mechanical loads. The criteria were calibrated and standardized so that the optimal coatings prototype should have the minimum value of each. The optimization task was therefore to find sets of solutions for which all criteria were simultaneously minimized. This, in the case of opposing criteria, leads to the formation of so-called Pareto-optimal sets. To analyze the selected feasible and Pareto-optimal solutions, methods were developed for selecting solutions and grouping them based on their potential operational properties. One of the methods based on the so-called utopian solution concept also made it possible to create a group of so-called compromise solutions, the main attribute of which was to have functional properties at an acceptable level previously defined by the decision-maker. To assess antimicrobial properties, experimental studies of bacterial population growth on ZrC coating surfaces were conducted, and the research was extended to titanium alloy substrates under specific laboratory conditions. To quantitatively analyze the obtained results, mathematical models were developed (using the Richards, Verhulst, and Gompertz equations) describing bacterial population growth over time. The models created will constitute new components of the previously developed coating optimization procedure, which will enable simultaneous optimization of mechanical and antimicrobial properties.

- Abstract:
Age-related macular degeneration (AMD) is a highly prevalent blinding disease of the elderly that affects both photoreceptors and the retinal pigment epithelium (RPE). Currently, a treatment option exists only for the less prevalent neovascular form of the disease. AMD is a multifactorial disease, with both modifiable risk factors such as diet and smoking, and non-modifiable risk factors such as age. As age causes several alterations in ocular tissues that lead to reduced oxygen delivery and hypoxic conditions in the retina, we use several mouse models of pseudo and true hypoxia to understand the pathology of the disease. Using histology, immunofluorescence, multi-omics as well as fluorescence lifetime imaging of genetically encoded biosensors we focus on the role of the hypoxia-inducible transcription factor HIF1 in the disease process. We show that the chronic activation of HIF1 downregulates oxidative phosphorylation and the Krebs cycle, which severely affects not only energy metabolism but also anabolism in photoreceptors leading to cell death and retinal degeneration. Since we have recognized HIF1 as the pathological HIF isoform in photoreceptors while others have defined overactivation of HIF2 as being causative for the degeneration of the RPE, we developed an AAV-based dual-acting RNA interference gene therapy strategy to simultaneously downregulate HIF1 in photoreceptors and HIF2 in the RPE. The application of the therapeutic virus significantly protected photoreceptors and retinal morphology in two models for more than one year post treatment. Our work suggests that targeting HIF transcription factors may be a promising strategy to preserve vision in patients affected by AMD.

Raman spectroscopy has become a cornerstone of biophotonics, oTering label-free molecular fingerprints with direct clinical relevance. Its convergence with artificial intelligence (AI) and complementary optical modalities is driving a paradigm shift from experimental method to clinically viable technology.
This presentation outlines our translational strategy along three representative examples. First, a high-throughput Raman blood analysis platform demonstrates how AI-based spectra interpretation enables rapid patient stratification, therapy monitoring, and culture-independent pathogen detection—essential for managing sepsis and antimicrobial resistance. Second, the regulatory-compliant Raman invaScope illustrates how real-time, AI-assisted endoscopic measurements support intraoperative tumor diagnostics, facilitating margin assessment in bladder as well as head and neck surgery. Third, multimodal endoscopic probes combine spontaneous and coherent Raman techniques with nonlinear optical imaging (SHG, TPEF) to generate morphochemical maps at subcellular resolution. Integrated with femtosecond-laser ablation, these probes open the path toward “seek-and-treat” workflows that unite diagnosis and therapy in a single system.
Underlying all approaches is a seamless diagnostic pipeline that spans sample preparation, spectral acquisition, and automated AI-based decision support. This integration minimizes operator dependency and enhances reproducibility, paving the way for robust deployment in diverse clinical settings.
We will conclude with an outlook on translational infrastructures and partnerships required to bring these Raman-based innovations into routine practice. Together, AI and advanced photonics are establishing Raman spectroscopy as a key enabler of intelligent, personalized healthcare.

This talk discusses how fluorine can be used to improve our work in developing and using chemically-modified oligonucleotides for downstream applications. First, I will discuss our work in using an ortho-functionalized tetrafluorinated azobenzene within short interfering RNAs (siRNAs) to optically control gene-silencing in both in vitro and in vivo systems. This will be followed by our work in studying gene activation, and how short activating RNAs (saRNAs) containing a 2'-fluoro adenosine in conjunction with other modifications such as a C3-propyl spacer can improve both resistance to nucleases and gene activation potential. Finally, I will present some data on how a fully 2'-fluorinated oligonucleotide (F-all) bound to a biopolymer, alginate, can be used to improve a prototype biosensor for forensic analysis.

- Abstract:
Since the pioneering work of Mourou [1] and Zewail [2], ultrafast electron diffraction (UED) has become the method of choice to study femtosecond structural dynamics in photoexcited matter, reaching picometre spatial and 100–240 fs temporal resolution in gas-phase and condensed systems [3–6]. Compact 100 keV UED instruments offer attractive simplicity and accessibility compared to facility-based MeV sources, but their performance is fundamentally constrained by space-charge effects [6]. For example, weakly relativistic 100 keV electron beams (0.55c) of high bunch charges (~20 fC) are often employed to achieve sufficient scattering signals at 1 kHz repetition rates, leading to space-charge broadening of electron pulses beyond the sub-picosecond regime. Even with radiofrequency (RF) [7] or THz compression [8], high-charge operation at 1 kHz limits pulse durations to ~150 fs, while single-electron operation at high repetition rate sacrifices throughput. Highly relativistic 3 MeV beams (0.99c) reduce space-charge effects by ~1,000 to achieve 29 fs compressed pulses [3], but their RF-driven accelerator source restricts it to 1 kHz operation, long beamlines (5-8 metres), indirect detection, and limited beamtime access.
We present a high-brightness, high-throughput 90 keV UED platform that overcomes these limitations. Using an RF compression cavity synchronized to the drive laser with <6 fs FWHM precision [9], we compress 0.4 - 7.4 ps electron pulses down to 91 - 114 fs, corresponding to a >65 compression factor, with charges ranging from 100 to 35,000 electrons per pulse, as confirmed with THz electron streaking [8]. Operating at 40 - 100 kHz with direct electron detection, our instrument combines femtosecond pulse durations with unprecedented throughput in the keV regime. Compared to state-of-the-art keV systems (>16 aC), our beam achieves shorter compressed durations (91 fs .vs. 100 - 150 fs) with higher brightness, while exceeding both keV and MeV UED in usable throughput by orders of magnitude.
In our recent work [10], we also detected time-resolved scattering signals with single-electron pulses and resolved electron-phonon dynamics in aluminium at 30 kHz using uncompressed beams. Here, we extend these capabilities into the high-charge regime via RF compression, combining high brightness (a per-pulse metric) with high throughput (a per-second metric). With high repetition rates, robust normalization to the measured unscattered beam, and direct detection, our instrument delivers among the brightest relativistic electron pulses and orders-of-magnitude higher usable throughput in the world, enabling high-sensitivity UED of gases, nanostructures, and photoinduced phase transitions.

We present computational results of many-body dispersion (MBD) interactions for 40 variegated pairs of molecular and atomic species: hydrocarbons, hydrosilanes, corresponding fluorinated derivatives, pairs which have multiple H---H interactions between the molecules, as well as pairs having p-p interactions, and pairs of noble gases. The calculations, reveal that the MBD stabilization energy (EDISP,MBD) obeys a global relationship, which is gravitational-like. It is proportional to the product of the masses of the two molecules (M1xM2) and inversely proportional to the corresponding distances between the molecular centers of mass (RCOM-COM) or the H---H distances (RH-H) of the atoms mediating the interactions of the two molecules. This relationship reflects the interactions of instantaneous dipoles, which are formed by the ensemble of bonds/atoms in the interacting molecules. Using the D4-corrected dispersion energy (EDISP,D4), which accounts for three-body interactions, we find that the EDISP,MBD and EDISP,D4 data-sets are strongly correlated (Figure 3). Based on past valence-bond modelling (Ref. 14), the dispersion interactions transpire primarily due to the increased contributions of the oscillating-ionic VB structures which maintain favorable electrostatic interactions, namely [Sub—C+ :H– +H :C– —Sub] and [Sub—C:– +H :H– C+—Sub]. This augmented contribution is attended by simultaneously diminished-weights of the destabilizing pair of structures, [Sub—C+ :H– –:H C+—Sub] and [Sub— :C– H+ +H :C– —Sub]; Sub symbolizes general residues. The local charges are propagated to tßhe entire ensemble of bonds/atoms by partially charging the Sub residues, thus bringing about the “gravitational-like” dependence of dispersion.

Artificial intelligence (AI) and machine learning (ML) have become important tools for chemists, enabling the analysis of vast chemical datasets. A key to harnessing these methods lies in translating molecular information into computationally useful representations, which are critical for training robust ML models. [1] We introduce a novel molecular fingerprinting method based on persistent homology, a concept from topological data analysis that captures essential topological features of molecular structures across multiple spatial resolutions. [2] This approach provides a unique perspective on molecular geometry, enhancing both the accuracy and interpretability of computational models. In this talk, I will outline the theoretical foundations of persistent homology and its adaptation for molecular applications. I will also highlight recent advancements that improve its accuracy and portability. Representative applications related to the discovery of novel CO2-philic molecules for carbon capture, [2] the design of ligands for catalyst optimization [3] and actinide capture, [4], polymer informatics, where topological fingerprints facilitate materials discovery, [5] and drug design [6] will be provided. This framework exemplifies the synergy between topology and AI, offering new avenues for chemical discovery and advancing our understanding of molecular systems.

However, along with changes in the environment, lifestyle, and other factors, eye-related diseases expand tremendously among the modern population. Although extensive research, there remains no method to fully reverse the already caused damage. The proposed strategy utilizes optogenetic technique to deliver the therapeutic cargo (potentially through the introduction of a light-sensitive channel) with the use of viral vectors to precisely targeted cells; however, this alone may not be sufficient to achieve a functional effect. Therefore, the broader objective is to identify an appropriate response amplifier capable of converting remaining retinal cells into synthetic photoreceptors, thereby enhancing the vision restoration.

Kenji Shoji is a translational medicine leader with a PhD in Physiology and over 19 years of experience in oncology R&D across academia and industry. His expertise spans from early-stage drug development to clinical trials, with a focus on innovative approaches in cancer research. As a postdoctoral researcher (2012-2015) at IRSET-INSERM U1085 (Rennes, France), he specialized in preclinical candidate characterization for cancer research. At Galapagos NV (2016-2022), he led a team in target discovery using AI, developed a flow cytometry platform, and introduced cutting-edge technologies like single-cell analysis and spatial transcriptomics. Currently (since 2022), as Biology Director at Oncodesing Precision Medicine, he oversees novel target validation, clinical biomarker selection, and preclinical package consolidation. He also leads the ONCOSNIPE project, a clinical study on various carcinomas aimed at discovering novel oncology targets.

Abstract: Biocatalysis is emerging as a key enabling technology for the development of a greener and more efficient chemical industry. Because natural enzymes are seldom suitable for direct use in chemical processes, however, protein engineering is typically required to optimize their properties for practical applications. For many desirable chemical transformations, natural enzymes may not even exist. Recent advances on the computational design of atomically accurate protein structures, combined with high-throughput evolutionary optimization, are providing a potential roadmap for reliably creating new-to-nature biocatalysts in response to diverse societal demands. In this lecture, the strategies, opportunities, and challenges facing this emerging field will be surveyed.

- Abstract:
Our laboratory at the Centre for Targeted Protein Degradation (CeTPD) reveals molecular information on protein interactions and ubiquitination complexes and mechanisms to design novel molecular therapeutics and concept. Protein degrader, also known as PROTACs (PROteolysis-TArgeting Chimeras) and molecular glues, recruit a target protein to a ubiquitin E3 ligase for targeted protein degradation. Formation of a stable ternary complex between the degrader, the E3 ligase and the target is a critical step that leads to productive tagging of the target protein by ubiquitination, and subsequent proteasomal degradation. Our research has illuminated fundamental structural and biophysical insights into molecular recognition and mechanism of action of protein degraders, that have guided the design and optimization of novel small molecules for hard-to-target proteins, including Brd9, SMARCA2, KRAS and LRRK2. In my lecture, I will illustrate key lessons from these fundamental work and drug discovery campaigns. I will then outline recent studies involving creative design and mechanistic insights, that have ushered development of exciting new concepts, such as trivalent PROTACs, dynamics combinatorial PROTACs, and intramolecular bivalent glues.

Since the seminal publication of Zyss et al. octupolar molecules with three-fold rotational symmetry (D3h or D3) have attracted sustained interest in the field of nonlinear optics due to their potentially large second- or third-order nonlinearities, making them often good two-photon absorbers,1-2 as is the case for N,N’,N”-isocyanurates (or 1,3,5-triazine-2,4,6-triones).3a-b We have previously shown that this is even more true when metal centers are incorporated as organometallic endgroups into their structure, but at the expense of their luminescence.4a-b In this presentation, we will now stick to organic derivatives and show, after recalling some of our initial results,3a how various structural variations can affect the photophysical properties of interest of these derivatives. We will also show through selected examples how this knowledge can be used for designing more efficient two-photon dyes or photosensitizers based on this old family of molecules. Comparison with closely related octupoles will also be made to show the specificities of these compounds.

Prof. Ewa Szczurek is the co-director of the Institute of AI for Health at Helmholtz Munich, Germany (from Feb 2024), and leads joint labs at Helmholtz Munich and at the Faculty of Mathematics, Informatics and Mechanics of the University of Warsaw, Poland. She was a visiting associate professor at Northwestern University in the United States (2023) and a visiting fellow at the Center for Interdisciplinary Research, Bielefeld, Germany (2016). She holds Master degrees in computer science from the University of Warsaw, Poland and Uppsala University, Sweden. She obtained her doctoral degree from the Max Planck Institute for Molecular Genetics in Berlin (2011), followed by a postdoctoral fellowship in Switzerland at ETH Zurich. Prof. Szczurek was a recipient of the distinction, scientific and didactic awards from the Rector of the University of Warsaw, as well as ETH Zurich and IMPRS fellowships for her postdoctoral and doctoral research. She acts as a program committee member for the ISMB and RECOMB-CCB conferences, area chair for NeurIPS, as well as associate editor for Genome Biology. Her research focuses on artificial intelligence, in particular probabilistic graphical models and deep generative models, and their applications in computational medicine. Her specific applications include oncology, pulmonology and the AI-driven design of antimicrobial peptides, a work for which she was recently awarded the ERC Consolidator grant.
Generative AI emerged as a promising technique to revolutionize drug discovery. The key challenge for applications of generative AI for molecule design is to equip the methods with controllability mechanisms, an area our lab has been recently focusing. Here, I will describe two approaches for AI-driven antimicrobial peptide design, namely a conditional variational autoencoder-based model, called HydrAMP, and a controlled diffusion model OmegAMP. I will also introduce our transformer-based joint generative and predictive model for molecule generation, Hyformer. Finally, I will go through Prospero, our recent active-learning and generative AI-based method for protein optimization. I will summarize promises and challenges that remain to be addressed in the field.

In ophthalmic research and practice, psychophysical and electrophysiological methods are used to obtain information on visual function. Psychophysical tests are subjective as they rely on the answers from the subject detecting and visually processing the light stimulus. Visual electrophysiology tests measuring changes in electric potentials originating from retinal neurons are objective but also difficult to conduct due to the requirement for the installation of special electrodes on the subject. This talk will focus on optoretinography, a new, objective, non-contact optical method for measuring photoreceptors’ activity.

Simulating electron transfer at reactive solid-liquid interfaces under constant electrochemical potentials of the constituents (electrons, ions, solvent etc.) is crucial to understanding the formation, functional and failure of electrochemical devices, while it is largely still a dream to fulfill. Albeit being sufficiently accurate in decreasing breaking and formation of chemical bond at solid surfaces, existing methods based on Kohn-Sham density functional theory (DFT) are unsatisfactory in system consistency, namely, simulating the open solid-liquid interface under grand-canonical conditions, as well as in scaling up the simulation due to its high computational cost. Herein, to improve the system consistency and computational efficiency, we develop density-potential functional theoretic (DPFT) schemes out of Kohn-Sham DFT, drawing upon ideas of orbital-free DFT, frozen density embedding theory, and tight-binding DFT. The DPFT transforms an all-atom, Kohn-Sham DFT description of the nonreactive electrolyte solution into a coarse-grained, field-based description, while retaining a Kohn-Sham DFT description for the reactive subsystem. In the absence of surface reactions, the solid electrode can be described using orbital-free DFT, reducing the computational cost further. On the conceptual level, the physical meaning of potential in DPFT is examined. Recent applications of DPFT to simulate EDLs with mesoscopic roughness and EDLs at supported nanoparticles will be introduced.

- Abstract:
Interfaces are important areas where reactions are taking place or where interactions on the molecular level determine the energetics. However, often it is difficult to obtain molecular level information from an interface, as the surrounding bulk molecules overwhelms the signal from the few interfacial molecules. In this talk we will discuss how sum frequency generation spectroscopy (SFG) can be used to obtain molecular level information of interfaces. Sum-frequency generation (SFG) spectroscopy is a technique capable of retrieving vibrational information of selectively interfacial molecules even if bulk amounts of liquid are present. Here, we will discuss two recent highlights of our work.
In the first part of the talk we will aim to understand the molecular scale interactions of a switchable emulsifier comprising poly(methacrylic acid) (PMAA) and poly(ethylene glycol) methacrylate (PEGMA). This emulsifier forms stable oil/water emulsions at pH > 10, but the emulsion becomes unstable at pH < 3 due to droplet aggregation. Using SFG, we study the PMAA/PEGMA polymer deposited on an aqueous subphase. We find that at pH 2.5, both MAA-EG H-bonds and MAA = MAA cyclic dimers contribute to droplet aggregation in the emulsion. In contrast, at pH 11.5 deprotonation and solvation of the PMAA component lead to a polymer restructuring forming a negative shell around the droplets, electrostatically stabilising the emulsion in alkaline conditions.
In the second part of the talk will follow the dynamics of water molecules at the TiO2-water interface after photoexcitation with UV light. A critical step in photocatalytic water dissociation is the hole-mediated oxidation reaction. Using femtosecond time-resolved SFG spectroscopy we could obtain molecular-level insights into the mechanism of this complex reaction under realistic conditions. Following the generation of holes at the surface immediately after photoexcitation of the catalyst with UV light, water dissociation occurs on a sub-20 ps timescale. At both pH 3 and 11, we observe the conversion of H2O into Ti-OH groups and the deprotonation of pre-existing Ti-OH groups. This study provides unique experimental insights into the early steps of the photo-induced dissociation processes at the photocatalyst-water interface.
Ellen Backus obtained her PhD in 2005 at Leiden University in the Netherlands. After a PostDoc at the University of Zurich, she moved back to the Netherlands for an independent PostDoc position at AMOLF in Amsterdam. In 2012, she joined the Molecular Spectroscopy department at the Max Planck Institute for Polymer Research as a group leader. Since 2018 she is professor for physical chemistry at the University of Vienna and leading the Ultrafast and Nonlinear Spectroscopy group. The overall research goal of her group is understanding the structure, relaxation dynamics, and reactivity of soft matter at interfaces and in bulk on the molecular scale.
