Description
Selective and sensitive chemosensing is crucial for numerous aspects of modern civilization and technology. Within this Project, we propose to apply redox active polymers to enhance electrochemically assisted SPR signal. This will result in the elaboration of a new opto-electrochemical transduction method. Moreover, the aforementioned polymers will contain receptor parts for selective recognition of insulin, peptide C (a side product of insulin production), and glimepiride (anti-diabetic drug). For that purpose, different approaches to molecular recognition in synthetic polymeric receptors will be applied.
According to L. C. Pauling, a two Noble Prize winner, "the secret of life is molecular recognition; the ability of one molecule to "recognize" another through weak bonding interactions". Molecularly imprinted polymers (MIPs) constitute example of tailor-made smart materials that can mimic recognition of biological receptors. However, MIPs outperform biological receptors in terms of durability, chemical stability, and production costs. Synthesis of these polymeric receptors consists of three steps, namely (i) assembling monomers around template molecule, (ii) polymerization in the presence of cross-linking monomer, and (iii) template removal. It results in the formation of molecular cavities in the polymer matrix that fit the target analyte molecules with shape, size, and pattern of complementary interactions, i.e., hydrogen bonds, electrostatic attraction, and π-π stacking. Therefore, these cavities can recognize and bind selectively target analyte molecules even in the presence of other similar ones in the tested sample.
Numerous examples of selective electrochemical and optical surface-plasmon resonance (SPR) sensors have been reported so far. To provide high selectivity, their surface had been coated with receptors of biological origin (biosensors) or synthetic receptors (chemosensors), including MIPs. They were employed in numerous fields including clinical diagnosis, cancer biomarkers detection, forensic sciences, warfare-agents detections, environmental analysis and food quality control. However, combining electrochemical measurement with SPR spectroscopy is a novel and less explored field. In this Project, MIPs in form of thin films or nanoparticles (NPs) will be deposited on the surface of Au SPR chips and applied for target molecules, peptide C, and glimepiride selective sensing.
