* Optical microscopy at ultimate limit of a single molecule. * Exploration of nanoscale behavior of molecules in complex local environments e.g. - spectral dynamics originate from changes within the chromophore and in the chromophoric environments, -diffusional motions, -conformational dynamics, -fluorescence resonant energy transfer, -Raman spectroscopy. * Influence of the environment on the efficiency of detection a single  fluorophore. * Imaging 3-dimensional orientation of single fluorescent objects

    Fluorescence patterns from differently oriented single Nile-blue molecules excited with radially polarized light
